Increased activity of alcohol oxidase at high hydrostatic pressure

Alcohol oxidase (AOx) from P. pastoris has potential applications in the production of carbonyl compounds and for detection quantification alcohols. However, AOx’s poor stability low activity have hindered its practical application. There are two fractions AOx with different thermal stability. High hydrostatic pressure (HHP) increased labile (L) + resistant (R) combined but not R fraction alone. The L 2.4-fold at 160 MPa 30 °C compared to 0.1 MPa. At higher temperatures, increase was greater due stabilization activation effects. reaction rate 50 17.9 ± 3.6 or 17.7 0.8 μM min−1 80 MPa, respectively, significantly under same conditions (18.4 2.7 min-1). energy between (41.5 10.5 kJ mol-1) (43.8 7.8 mol-1). HHP enables use enzyme little loss an catalytic rate.
